==Mechanism of Action==

Novobiocin works by inhibiting bacterial DNA gyrase by acting as a competitive inhibitor of the ATPase reaction performed by the GyrB subunit of the gyrase enzyme.


==Spectrum of Activity==
It has a narrow spectrum mainly targeting gram-positive species.
* Very active against ''S.aureus''. It works less well against streptococci and fastidious gram-negatives.
* It is at its least active against enterbacteriaceae and ''Pseudomonas''.
* It works synergistically with [[Penicillins | Penicillin G]] against ''Staphs'' and ''Streps''. They are often combined in intramammary preperations, this is Novobiocin's only use in Veterinary medicine.


==Pharmacokinetic Considerations==
It isn't used systemically but only topically with other drugs to treat mastitis.
