Novobiocin is a coumarin antibiotic produced by Streptomyces
Mechanism of Action
Novobiocin works by inhibiting bacterial DNA gyrase by acting as a competitive inhibitor of the ATPase reaction performed by the GyrB subunit of the gyrase enzyme.
Spectrum of Activity
It has a narrow spectrum mainly targeting gram-positive species.
- Very active against S.aureus. It works less well against streptococci and fastidious gram-negatives.
- It is at its least active against enterbacteriaceae and Pseudomonas.
- It works synergistically with Penicillin G against Staphs and Streps. They are often combined in intramammary preperations, this is Novobiocin's only use in Veterinary medicine.
Pharmacokinetic Considerations
It isn't used systemically but only topically with other drugs to treat mastitis.
Side Effects and Contraindications
Some cows have experienced skin eruptions on the udder after use.
